He was born Darren Lee Sproles to Larry Sproles and his wife Annette (died in 2004) on 20 June 1983. His father was playing for MidAmerica Nazarene University as a running back. He commenced his education at the Oregon Trail Junior High before going to Olathe North High School. He was a star in football since his high school days, and he earned several titles for his prowess. He also was active in track and field at Olathe North. He made his college debut in the 2001 season against New Mexico State, and he finished the season with 28 carries for 210 yards. He was drafted during the 2005 NFL Draft by the San Diego Chargers as 130th overall pick. He spent his first season as the third-string back behind Michael Turner, LaDainian, and Tomlinson. The following season was not very favoring for him because he had an injury and did not play.
He returned in the 2007 season, and immediately, he made history by returning a kick-off and a punt for his NFL touchdown in a match where the Charges defeated Indianapolis by 23-21.
