Leslie Claypool was born on September 29, 1963, in Richmond, California, to a working-class family whose profession was of car repairing. He decided to become a bass player, pursuing the instrument from the age of fourteen.

Claypool has cited Chris Squire, Tony Levin, Roger Waters, Geddy Lee, Paul McCartney, Geezer Butler, Bootsy Collins, John Paul Jones, and the Residents as most of his musical influences. He went to school with and was a friend of Kirk Hammett of Metallica.
